To compare CB2R binding, β-Caryophyllene has the highest CB2 cannabinoid activity.  Cannabinol (CBN) binds to the CB2 receptors as a partial agonist with a binding affinity of CB2 Ki at 126.4 nM[9] while Delta-9-Tetrahydrocannabinol binds to the CB2 receptors as a partial agonist with an affinity of Ki at 36nM.[10]

The endocannabinoid system plays a major role in the function of the human body, regulating eating habits, sleep, and mood. It also controls pain and pleasure, in addition to inflammation. When to take BCP+ 30x Liquid Extract Cannabinoid Oil

Because oils are slow-acting and long-lasting, it’s recommended that you start low, and go slow when experimenting with oils.

Place a few drops under the tongue to gauge your sensitivity to BCP+ Cannabinoid Oil.

When it’s held under the tongue, the BCP+ Cannabinoid Oil is absorbed by the mucous membranes and then directly into the bloodstream, bypassing the digestive tract, which allows for a quicker delivery.

Oils are a much more concentrated version of flowers and buds and can have some unpleasant side effects like nausea, dizziness, thirst, and a drop in blood sugar levels if you go too quickly.

Taking the oil with food in your stomach has been known to
minimize nausea.

Cannabinoids have been receiving more and more attention in the news lately and you may find yourself wondering: what is the endocannabinoid system and what role does it play in the human body?

The endocannabinoid system works to keep the human body in balance.

In order to understand the endocannabinoid system, it’s also important to understand the key biological concept of homeostasis, which refers to any process or reaction that the body utilizes to actively maintain a stable internal environment. In many cases, these functions geared towards maintaining balance are necessary for any organism to survive.

Disturbances to physiological equilibrium, if uncorrected, can lead to impaired health, with symptoms that range from headaches to more serious diseases like arthritis, epilepsy, stroke, obesity, Alzheimer’s disease, cancer, and glaucoma. The body’s ability to adjust to changes and maintain homeostasis directly impacts the state of health of that organism.

The endocannabinoid system is a molecular system responsible for regulating and balancing many processes in the body, including immune response, communication between cells, appetite and metabolism, memory, and more.

It is through this system of receptors and metabolic enzymes that cannabinoids interact with the human body and trigger beneficial effects.

Despite the integral role the endocannabinoid system has in human body function, it was discovered relatively recently and much of its importance is only just becoming understood by scientists and the medical community at large.

A Brief History of the Endocannabinoid System

In 1998, Vincenzo Di Marzo, one of the world’s leading cannabinoid scientists, stated that these physiological processes in the human body: mood, appetite, sleep, memory, and pain-sensation are in the domain of a biological regulatory system wired to keep the body in balance.

Named after the plant that led to its discovery, the endogenous cannabinoid system is perhaps the most important physiologic system involved in establishing and maintaining human health.

It is composed of endocannabinoids, which are endogenous lipid-based retrograde neurotransmitters that bind to cannabinoid receptors, and cannabinoid receptor proteins that are found throughout the body in the brain, organs, connective tissues, glands, and immune cells.

In each tissue, the cannabinoid system performs different tasks, but the goal is always the same: homeostasis or equilibrium. In fact, almost every physiological process is affected by the endocannabinoid system on some level. This means it can be targeted to treat many diseases.

What is surprising is that the endocannabinoid system was discovered less than 30 years ago.

In 1964, Dr. Raphael Mechoulam, successfully isolated , a non-psychotropic cannabinoid generally associated with antioxidant and neuroprotectant properties.

These cannabinoids are referred to as “phytocannabinoids” because they are naturally occurring in plants. Being able to isolate these phytocannabinoids for the first time was a critical stepping stone in discovering the endocannabinoid system.

The Cannabis plant contains beta Caryophyllene (BCP), a non psychoactive phytocannabinoid that activates the CB 2 Receptors in humans.

In 1990, a molecular biologist, Lisa Matsuda and her colleagues at the National Institute of Mental Health identified the endocannabinoid system.

Shortly thereafter, Mechoulam’s research led him to the discovery of two endocannabinoids: anandamide and 2-arachidonoylglycerol, or 2-AG.

Endocannabinoids are cannabinoids that are naturally produced in the body by the brain.

These endocannabinoids bind to cannabinoid receptors on target cells throughout the body, triggering a cellular response which is amplified or diminished as metabolic enzymes destroy or make more endocannabinoids.

This activity produces diverse effects that range from anti-inflammatory responses to euphoria.

These discoveries triggered an explosion of research exploring the endocannabinoid system. The system has been implicated in multiple physiological functions and we now have valuable knowledge about the pharmacology, biochemistry, and clinical effects of endocannabinoids.

It has been established that endocannabinoids have a role in the pathology of many disorders and it is believed that they also serve a “protective role” in many medical conditions.

Diseases like pain, inflammation, multiple sclerosis, anorexia, epilepsy, glaucoma, anxiety, depression, schizophrenia, cardiovascular disorders, cancer, obesity, metabolic syndrome related diseases, Parkinson’s disease, Huntington’s disease, Alzheimer’s disease and Tourette’s syndrome could possibly be treated by drugs modulating the endocannabinoid system. 

Cannabinoid Receptors: What They Are and What They Do:

CB2 receptors are predominantly distributed in the immune system. They appear to be responsible for anti-inflammatory and other immune-related activity.

The two primary endocannabinoids produced by the human body interact directly with these receptors. .

Cannabinoid Receptors You Probably Haven’t Heard About:

There is new evidence that suggests that the endocannabinoid system is more complex than first believed. Since 1999, we have learned that cannabinoids were still able to affect blood pressure, pain, inflammation, and gastric motility in the absence of CB1 and CB2 receptors.

This led to the discovery of at least eight other receptors to which cannabinoids bind. GPR18, GPR55, GPR119 have functions in the immune system, CNS, and gastrointestinal systems respectively, while others are capable of regulating pain transmission and cellular function.

Endocannabinoids bind to cannabinoid receptors on target cells throughout the body, triggering a cellular response which is amplified or diminished as metabolic enzymes destroy or make more endocannabinoids.

A case for the existence of a condition referred to as Clinical Endocannabinoid Deficiency?

Researchers believe that the underlying cause of many ailments, particularly those related to the immune system and inflammation, could be a disorder referred to as clinical endocannabinoid deficiency.

Endocannabinoid deficiency is a theory for the symptoms and conditions that develop when the body’s endocannabinoid system isn’t functioning properly. We’ve previously discussed the endocannabinoid system and its role in regulating a wide array of body processes to maintain our health.

The body naturally produces neuromodulatory lipids called endocannabinoids, which interact with cannabinoid receptors throughout the body to elicit chemical responses that keep the body in balance.

The theory of clinical endocannabinoid deficiency suggests that in some cases the body doesn’t produce enough endocannabinoids or enough receptors for the endocannabinoid system to function properly.

As a result, the many functions aren’t regulated properly and the body becomes unbalanced, allowing diseases to arise.

Renowned cannabinoid researcher Dr. Ethan Russo first proposed the theory of clinical endocannabinoid deficiency in 2004. In a study, published in Neuro Endocrinology Letters, Russo suggested that deficient cannabinoid levels might explain why some conditions develop.

Russo had examined the available literature and found evidence that people diagnosed with certain conditions had lower levels of endocannabinoids. He suggested that a person being deficient in endocannabinoids and/or cannabinoid receptors could explain why supplementing with cannabinoids found naturally present in some plants, (phytocannabinoids) were proving effective at alleviating some conditions.

“Migraine, fibromyalgia, [irritable bowel syndrome] and related conditions display common clinical, biochemical and pathophysiological patterns that suggests an underlying clinical endocannabinoid deficiency that may be suitably treated with cannabinoid medicines,” Russo concluded in his research review.

Cannabinoids found in plants (phytocannabinoids) are analogous to the endocannabinoids naturally produced by the body. Research shows that they interact with the cannabinoid receptors of the endocannabinoid system to elicit chemical responses that help bring about homeostasis. (equilibrium – balance)

A deficiency in endocannabinoids has been linked to several other disorders besides migraine, fibromyalgia and irritable bowel syndrome.

The endocannabinoid system is responsible for regulating the release of neurotransmitter, so diseases that are attributed to their dysfunction, such as Alzheimer’s disease and Parkinson’s disease, may be related to clinical endocannabinoid deficiency.

While more research needs to be done before endocannabinoid deficiency can be proven, the theory could open the door for methods to prevent and manage certain conditions.

Russo believes cannabinoids sourced naturally from plants, (phytocannabinoids) are essential nutrients that can minimize certain diseases by assisting the endocannabinoid system and ensuring that it functions properly.

BCP Cannabinoid beta Caryophyllene, THC and CBD are the most widely researched cannabinoids in Cannabinoid science. However, the reality is that these compounds are only part of the story.

beta Caryophyllene (BCP), along with at least other 113 other Cannabinoids, operate in sync with hundreds of other compounds to modulate BCP’s therapeutic effects when consumed and ultimately give BCP its versatility in providing relief for a multitude of medical ailments.

The term “entourage effect” refers to the concept that all the unique components of a plant work together in concert to exert a therapeutic effect greater than any single compound by itself.

A 1981 research study by scientists J.W. Fairbairn and Joan T. Pickens provided evidence confirming the existence of the entourage effect in Cannabinoids. Using the ring test, the researchers demonstrated that whole plant extracts produced 330% more activity than the isolated plant compound alone. They hypothesized that plant Cannabinoids contain “synergist” and “inhibitor” compounds – serving to enhance therapeutic effects while inhibiting potentially negative reactions.

How is this possible? Cannabinoid containing plants are complex, with hundreds of molecules and compounds, including cannabinoids, terpenes, flavonoids and other phytochemicals, each of which is thought to affect the brain and body in a different way.

These elements interact synergistically to produce and amplify the overall therapeutic benefits of Cannabinoids. The term “entourage effect” refers to this phenomenon with results that make the whole plant more beneficial than the sum of its individual molecules.

THE ENTOURAGE EFFECT

This notion that the compounds present in plant cannabinoids work in concert to produce stronger benefits than they would separately was first introduced in a 1998 Paper: An entourage effect: inactive endogenous fatty acid glycerol esters enhance cannabinoid activity by scientists Shimon Ben-Shabat and Raphael Mechoulam. The scientists were the first to establish that it is the synergistic relationship between these compounds that is largely responsible for the therapeutic effects of Cannabinoids.

Expanding on this in a 2009 paper“Synergy research: Approaching a new generation of phytopharmaceuticals,” scientists Hildebert Wagner and Gudrun Ulrich-Merzenich outlined and discussed the following basic mechanisms of the entourage effect:

  1. Whole-plant derived compounds can affect multiple targets within the body.
  2. Whole plant synergy improves the bioavailability, resorption and solubility rate of active compounds.
  3. Whole plant synergy assists in overcoming bacterial defense mechanisms.
  4. Whole plant synergy contributes to minimizing adverse side effects of active compounds.

From this research, it’s clear that the entourage effect supports the idea that whole plant medicine is superior to single isolated molecules.

THE TERPENE ASSIST

Ethan Russo, M.D., a neurologist who now serves as director of research and development at the International Cannabinoids Institute, published an article  in the British Journal of Pharmacology in 2011 Taming THC: potential cannabis synergy and phytocannabinoid-terpenoid entourage effects in which he details how plant compounds influence each other’s mechanisms. The article, was one of the first to examine how the entourage effect works when Cannabinoids are administered to mammals.

Russo found that terpenes – the fragrant plant oils that give plants their smell – are quite potent on their own, and provide a broad range of effects, including sedative, anti-bacterial, anti-inflammatory and palliative therapy. However, he also uncovered how terpenes seem to change the way in which Cannabinoids bind to their respective receptors, thus altering the way in which Cannabinoids take effect internally.

Stating that cannabinoid-terpenoid interactions “could produce synergy with respect to treatment of pain, inflammation, depression, anxiety, addiction, epilepsy, cancer, fungal and bacterial infections,” Russo suggested that further research into the entourage effect holds great potential for improved pharmaceutical solutions through plant phytochemistry.

WHAT DOES THIS MEAN FOR CANNABINOID-BASED THERAPIES?

The entourage effect implies that we’re not just relying on one thing – not just (BCP) Cannabinoid beta Caryophyllene for example, but rather, a combination of compounds. Because you are using a product that has more than one active ingredient, you are getting benefit from the entourage effect. Your result is greater than if you just used one of those compounds.”

Reflecting on this effect in conjunction with the current lack of an established standard of care in Cannabinoid dosing, it could be viewed as a positive opportunity for ownership of self-care.

It empowers the User, it gives the User ownership over their own care, self determining what they put into their body, & how they do so. They can choose their product and select their preferred method of delivery – sublingual is an option, as is an edible, vaping another, and so on. They are able to medicate with as much or as little as they need because it is a very safe holistic therapy – much safer than opioids.

Acknowledging that there is not currently a way to properly recommend exact dosage, it is incumbent upon Users to monitor their own results and the product, dosage and delivery method that caused the effect.

Despite these promising studies, the entourage effect is still hotly debated in scientific circles and there is a growing effort to prove its validity. This increased interest reflects changing attitudes the World over, about Cannabinoid use for medicinal self therapy.

The recent FDA approval of GW Pharmaceuticals’ Epidiolex for seizure management of patients with Lennox-Gastaut syndrome and Dravet syndrome for example, is another indication of the growing acceptance of cannabinouid-based therapies.

There is much left to be uncovered in the potential of plants that impact the Endocannabinoid System. Studying the entourage effect will be key to furthering our understanding of how the components of Plant Cannabinoids, all work in concert together. This could lead to the development of innovative plant based therapies tailored directly to specific User needs, and has the potential to revolutionize the medicinal Cannabinoid Industry.
